For safety and maintenance purposes, the access is allowed after training and only using special suit - bunny suit (see picture below). Its main purpose is to block emission of particles from humans into the cleanroom. But also it can protect the people working in the cleanroom against some dangerous chemicals.
Vacuum is created in chambers by pumps; vacuum is required for example for plasma discharges at low pressures.
